Hazard Analysis and Critical Control Points in Handling Frozen Tuna Steak (Case Study at PT. Awindo International, Jakarta, Indonesia)

This research aims to analyze the hazard and determine critical control points in the process of handling frozen tuna steak at PT. Awindo International located in Muara Baru, North Jakarta. The research was conducted at PT. Awindo International from March to April 2021. The research was conducted using the case study method. The research procedure includes observation of the handling process flow, analysis of potential hazards, identification of critical control points, organoleptic, microbiological and chemical testing. The data obtained from the study were analyzed descriptively. Hazards that have the potential to occur in the process of handling frozen tuna steak are biological, physical and chemical hazards. The results of the hazard analysis obtained critical control points at the stage of receiving raw materials and metal inspection. The results of the organoleptic test of raw materials as a whole have a good value and are in accordance with company standards and Indonesian national standards (SNI), so that they can proceed to the production stage. The results of the microbiological test showed that complied with the standards, namely Salmonella sp. negative, Vibrio cholerae negative, Escherichia coli <1.8 MPN/g, Coliform <1.8 MPN/g, ALT 12 x 103 colonies/g. The results of the chemical test showed good results with the value of the chemical content that complied with the standards, namely histamine 2.92 mg/kg, mercury 0.05 mg/kg, negative lead, and cadmium 0.019 mg/kg.

Macrozoobenthos Community Structure in Mangrove Forest Ecotourism Area Karangsong Indramayu Regency, West Java

This research aims to analyze the diversity of macrozoobenthos species and density in mangrove ecotourism waters Karangsong Indramayu Regency West Java. The research was conducted in West Java starting from January 2021 to March 2021. This research method uses primary data in the form of physical and chemical data to measure the density, diversity and uniformity of macrozoobenthos. Data retrieval technique uses Purposive Sampling in the determination of 3 stations and 4 repetitions. The results of the research found 15 species of Makrozoobenthos that live in mangrove ecosystems namely Telescopium telescopium, Balanus perforates, Clypeomorus zonata, Cassidula aurisfelis, Cypraea leucogaster, Gyrineum natator, Polinices powisianus, Perna viridis, Placuna placenta, Tellina sp, Gari sp, Meretrix meretrix, Dossinia sp, Saccostrea cucullate and Anadara transversa. The density of macrozoobenthos ranges from 294 – 548 ind/m2. The diversity index of macrozoobenthos at stations 2 and 3 is in the medium category with a value of 2.44 and 2.822 and the diversity at station 1 is in the high category with a value of 3.10. Uniformity values range from 0.84 – 0.891 with a high category. The relationship of Water Quality with Makrozoobentos has an R2 value of 90% water quality can affect the life of macrozoobenthos and as much as 10% is influenced by other factors.

Culture Pond Influences the Growth and Survival of Clarias gariepinus Fingerlings reared in Poly and Monoculture

Monoculture is the rearing of single species of fish in a pond while polyculture is the farming of two or more species in the same physical space. This study aims to compare the growth parameters of Clarias gariepinus fingerlings in monoculture and polyculture, using concrete and earthen ponds in a randomized complete block design. The polyculture, comprised of Clarias gariepinus and Oreochromis niloticus. 480 fingerlings were used; the stocking density was 40fish in a 3x2x1.5m depth with initial weight (4.84±0.11547) for C. gariepinus in monoculture and 40 each in polyculture with an initial pooled weight of O. niloticus (50g) replicated thrice, it was fed for eight weeks with commercial feed (40% crude protein). Weekly, random sampling was done to measure the growth and adjust the feeding rate. The results revealed that polyculture has the best mean final weight (160.07±0.18559g) and weight gain of 155.29, while the highest total length (45.666±3.179) was recorded in monoculture for Clarias gariepinus reared in an earthen pond. The polyculture had the highest specific growth rate (2.46), Relative growth rate (246.49), and condition factor (0.184). There was a significant difference (P<0.05) in monoculture of both concrete and earthen ponds.

Preservation of Squid (LOLIGO SP.) Using Yogurt in Low Temperature Storage on Organoleptic Characteristics

This research aims to determine the shelf life of squid (Loligo sp.) using Lactic Acid Bacteria in yogurt and its organoleptic characteristics. This research was carried out at the Fishery Product Processing Laboratory, Faculty of Fisheries and Marine Sciences, Padjadjaran University in November 2020. This research was carried out using yogurt as a preservative for squid (Loligo sp.) in the treatment of LAB concentrations used 0%, 1, 5%, 3%, 4.5%, and 6%. Observations were made for 11 days and stored at a temperature of 5-10oC, and organoleptic testing (discoloration, brilliance, aroma, and texture) was carried out on days 1, 3, 5, 7, 8, 9, 10, 11 for the treatment 1.5%, 3%, 4.5% and 6% and organoleptic testing on days 1, 3, 5, 7, 8 for control samples. The observed data were analyzed for pH and organoleptic with Friedman test and Chi-Square test. The results showed that 1,5% Lactic Acid Bacteria into yogurt as a preservative in squid has the best effect on the shelf life and organoleptic characteristics of squid during storage at low temperatures with an acceptable limit of up to 10 days.

Optimization of Hormone for Artificial Breeding and Seed Production of Clarias batrachus (Linnaeus, 1758) Under Captive Condition

Aims: The study aimed to develop brood stock of Magur (Clarias batrachus), artificial breeding and seed production under captive condition using growth hormones in north-eastern Bangladesh.

Study Design: Fifteen pairs of brood Clarias batrachus were used in this experiment. Brood fishes were reared up to maturation for spawning operation in the farm by providing artificial diet for 4-5 months before onset of breeding season. In the present experiment, the brood fishes were induced with three growth hormones viz., Ovupin (100 mg dompridone and 0.2 mg S-GnRHa) at the rate of 1 ml.kg-1 body weight, Flash (20 mg S-GnRHa, 10 mg dompridone IP and propylene glycol) at the rate of 1 ml.kg-1 body weight and Human Chorionic Gonadotropin (HCG) hormone at the rate of 2272 IU.kg-1 body weight; respectively. The broods were injected with single injection in all treatments.

Place and Duration of Study: The present study was conducted on induced spawning of Magur (Clarias batrachus) at the Reliance Aqua Farm, Boilor, Trisal of Mymensingh district from March to August, 2016.

Methodology: Sexually mature healthy, uninjured male and female fishes were collected from the broodstock ponds for induced breeding. The fishes were transferred in the clean and germ free conditioning tanks for about 6-7 hours. During conditioning, continuous splash of water was given by keeping the male and female separate. The fishes were then weighed individually and injected intramuscularly with Ovupin, Flash and HCG hormone respectively. Only females were injected at 45° angle of the caudal peduncle. About 23-24 hours of post-injection the testes from males were dissected and mixed with stripping eggs for fertilization.

Results: The incubation period for fertilized eggs were 23-24 hours for all the treatments. Fertilization rates were 78.20±0.52, 93.09±0.61 and 81.60±0.97 % and hatching rates were 54.93±0.61, 75.77±0.47 and 64.41±0.71 % in Ovupin, Flash and HCG treatments, respectively.

Conclusion: This study revealed higher fertilization (93.09 %) and hatching (75.77 %) rate was achieved with Flash hormone. Therefore, Flash hormone could be effective for the artificial breeding and seed production of Magur (Clarias batrachus).
